The serpin plasminogen activator inhibitor-1 (PAI-1) is afast and specific inhibitor of the plasminogen activatingserine proteases tissue-type and urokinase-type plasminogenactivator and, as such, an important regulator in turnover ofextracellularmatrixand infibrinolysis. PAI-1 spontaneouslyloses its antiproteolyticactivityby inserting its reactive centreloop (RCL) as strand 4 in b-sheet A, thereby converting tothe so-called latent state.
