Three mammalian ADAMTS enzymes, ADAMTS-1, -4and -5, are known to cleave aggrecan at certain glutamylbonds and are considered to be largely responsible for car-tilage aggrecan catabolismobservedduring the developmentof arthritis. We have previously reported that certain cate-chins, polyphenolic compounds found in highest concen-trationingreentea(Camellia sinensis), are capable ofinhibiting cartilage aggrecan breakdown in anin vitromodelof cartilage degradation.We have nowclonedand expressedrecombinant human ADAMTS-1, -4 and -5 and reporthere that the catechin gallate esters found in green teapotently inhibit the aggrecan-degrading activity of theseenzymes, with submicromolar IC50 values....
