This study aimed to determine the effects of an herbal formula named TD0014 on the electrostimulationinduced erection in a rat model. Eighteen sixteen-week-old male Wistar rats were used. The intracavernous pressure and the arterial blood pressure were simultaneously monitored during cavernous nerve electrical stimulation before and after the administration of TD0014 (1.8 g/kg).

Statistical analysis wasperformed on measurements of maximal intracavernous pressure (MIP), mean arterial blood pressure(MAP) and the MIP/MAP. The administration of TD0014 resulted in a significant increase in the baselinelevel of intracavernosal pressure (ICP) before stimulation in comparison to the control group (P < 0.05).The findings also showed that the maximal ICP and the area under the curve (AUC time × ICP curves)in TD0014 group were greater than distilled water group, however, this difference was not significant.TD0014 had no significant effect on the mean arterial pressure. These results indicated that the herbalformulation TD0014 may have positive and selective effects on improving erectile functions in male rats.Keywords: TD0014, cavernous nerve stimulation, intracavernous pressure, ratI. INTRODUCTIONErectile dysfunction (ED) is a termrecommended by a panel of experts in1992 to replace the term “impotence” [1].ED is when a man is unable to get and/orkeep an erection. It is not a disease, buta symptom of some other problem, eitherphysiological, psychological or a mixtureof both [2]. Although physiological EDitself is not life threatening, it is a strongpredictor of high-mortality diseases such ascardiovascular disease [3]. Recentlydeveloped phosphodiesterase type-5 (PDE5) inhibitors have been widely used as firstline therapeutics to treat ED [5]. PDE5 isan enzyme found primarily in the smoothmuscle of the corpus cavernosum thatselectively cleaves and degrades cGMPto 5′-GMP. PDE5 inhibitors are similar instructure to cGMP; they competitively bindto PDE5 and inhibit cGMP hydrolysis. Theincreased amounts of cGMP enhance theeffects of NO a potent vasodicator. Thisincrease in cGMP in the smooth muscle cellsis responsible for prolonging an erection [6].Although large, multicenter clinical trialshave shown the efficacy and tolerability ofJMR 111 E2 (2) - 2018JOURNAL OF MEDICAL RESEARCHthese drugs in patients with ED of variousetiologies and a broad range of severity;however, 30% to 35% of patients fail torespond. The use of PDE - 5 inhibitors mayresult in several side effects, including visualdisturbances, headache, facial flushing,rhinitis, and indigestion. Other treatmentsfor ED include penile injection therapy orpenile implants. However, such methodsare invasive and irreversible, and aretherefore not widely used [7]. Thus, thereis a continued need for the development ofnew noninvasive and effective therapies totreat patients with ED.Despite the remarkable developmentsof modern medicine, many people arestill favorably disposed towards herbalmedicines owing to the aggressivetreatment protocols, toxicity, and drugtolerance associated with modern therapies.The widespread use of herbal medicines,however, requires scientific verification oftheir effects.TD0014 is a preparation whichcomprises of thirty-three medicinal plants.The composition of TD0014 has severalmedicinal herbs that have been studiedand used since ancient times in traditionalfolk medicine as an aphrodisiac [8 - 15].However, no studies have shown reliableevidence of their effect on reproductivefunction, or toxicity when taken in TD0014.Therefore, the purpose of this study was toexplore the impact of TD0014 on erectilecapacity in adult male rats.II. MATERIALS AND METHODS1. MaterialsJMR 111 E2 (2) - 2018Herbal formula TD0014 preparationTD0014 was manufactured as hard pillsaccording to the quality standard of SunstarJoint Stock Company, Vietnam.
