3 hiệu quả bảo quản tế bào gốc tủy răng của dung dịch chuyên chởNghiêncứuYhọcYHọcTP.HồChíMinh*Tập18*PhụbảncủaSố1*20143HIỆUQUẢBẢOQUẢNTẾBÀOGỐCTỦYRĂNGCỦADUNGDỊCHCHUYÊNCHỞLêHoàngSơn*,NgôThịQuỳnhLan*,TrầnLêBảoHà**TÓMTẮTMụctiêu:Nghiêncứunhằmđánhgiáhiệuquảbảoquảntếbàogốc(TBG)tủyrăngbằngdungdịchchuyênchởmới,khôngđắttiền,phachếdễdàngvớicácthànhphầnphổbiếnởViệtNam.Vật liệu và phương pháp nghiên cứu: Dung dịch chuyên chở được pha bằng cách hòa tan 3 ốnggentamycin80mg/2mltrong500mldungdịchnướcmuốisinhlý.Nghiêncứuthửnghiệmđượcthựchiệntrên43răngkhônnguyênvẹnthuthậptừcácbệnhnhân18–35tuổiđếnnhổrăngtạiKhoaRăngHàmMặt.Răngđượcloạibỏmômềm,tạorãnhsâu2mmtrênthân–chânrăngvàbảoquảnbằngdungdịchchuyênchởtrong6giờ,9giờ,12giờởđiềukiện4°C.Đánhgiámôtủynhiễmtrùngkhimôitrườngnuôicấyvẩnđụcsau3–5ngàynuôicấyvàmẫumôtủyxemnhưđãchếtkhikhôngcótếbàopháttriểnsau2tuầnnuôicấy.Kếtquả:Trong43răngkhôn,sốlượngrăngđượcbảoquảntrong6giờ,9giờvà12giờlầnlượtlà14răng,16răngvà13răng.Có1mẫutrongnhóm12giờbịnhiễmkhuẩnsau5ngàynuôicấy.Tỉlệmẫukhôngnhiễmđạt100%ởnhóm6giờvà9giờ;92,3%ởnhóm12giờ.Tỉlệmẫunuôicấychotếbàobámdínhlà92,9%đốivớinhóm6giờ;75,0%đốivớinhóm9giờvà53,8%đốivớinhóm12giờ.Kếtquảflow‐cytometrychothấyquầnthểtếbàoP4đượcphânlậpdươngtínhcao(>99,5%)vớicácmarkercủaTBGtrungmôvàbiểuhiệnkháthấp(7,5%‐10,6%)vớicácmarkertếbàotạomáu.Kếtluận:Dungdịchchuyênchởđềnghịcóthànhphầndễtìm,phachếđơngiảnvàkhôngđắttiềnnhưngvẫncóhiệuquảcaotrongviệcbảoquảnsựsốngcủaTBGtủyrăng.CácmẫutủyrăngvẫncóthểchoTBGtrungmôsau12giờbảoquảnvàhiệuquảbảoquảntươngđươngvớicácdungdịchthươngmạitrong6giờ.Từkhóa:tếbàogốctủyrăng,dungdịchchuyênchở,flowcytometry,tếbàogốctrungmô,markerABSTRACTTHEEFFICIENCYOFANOVELTRANSPORTSOLUTIONINPRESERVINGDENTALPULPSTEMCELLSLeHoangSon,NgoThiQuynhLan,TranLeBaoHa*YHocTP.HoChiMinh*Vol.18‐SupplementofNo1‐2014:282‐287Objectives: The purpose of this study is that testing the efficiency of a suggested transport solution inpreservingdentalpulpstemcells(DPSCs)in6,9,and12hoursaftertheteethwereextracted.Materials and Methods: A new transport solution was fomulated by mixing three jars of gentamycin80mg/2mL(inliquidform)with500mlsterilesaline.Thesolutionwaspouredintoseveralsmallvials.Forty‐threeextractedintacthumanthirdmolarswereobtainedfromOralSurgicalCenterattheFacultyofOdonto–Stomatology.Theteethweredividedintothreegroups:groupA(n=14)–storedin6h,groupB(n=16)–storedin9h and group C (n=13) – stored in 12h. Teeth were externally sterilized and DPSCs were isolated using theprotocoladaptedfromStemCelllaboratoryattheUniversityofSciences.Results:Thepercentageofuncontaminatedsampleswas100%forgroupAandB,92.3%forgroupC.The*Khoa RHM – Đại học Y Dược TP.
