Lysophosphatidicacid (LPA), oneof thenaturallyoccurringphospholipids, stimulates cellmotility through theactivationof Rho family members, but the signaling mechanismsremain tobe elucidated. In thepresent study,we investigatedthe roles of p21-activated kinase 1 (PAK1) onLPA-inducedfocal adhesion kinase (FAK) phosphorylation and cellmotility. Treatment of human melanoma cells A2058 withLPA increased phosphorylation and activation of PAK1,which was blocked by treatment with pertussis toxin andby inhibition of phosphoinositide 3-kinase (PI3K) with aninhibitor LY294002 or by overexpression of catalyticallyinactive mutant of PI3Kc, indicating that LPA-inducedPAK1 activation was mediated via a Gi protein and thePI3Kcsignalingpathway....
