In the leukemic macrophage cell-line THP-1, a fraction ofthe secreted matrix metalloproteinase 9 (MMP-9) is linkedto the core protein of chondroitin sulfate proteoglycans(CSPG). Unlike the monomeric and homodimeric formsof MMP-9, the addition of exogenous CaCl2 to theproMMP-9/CSPG complex resulted in an active gelatinasedue to the induction of an autocatalytic removal of theN-terminal prodomain. In addition, the MMP-9 wasreleased from the CSPG through a process that appearedto be a stepwise truncation of both the CSPG core proteinand a part of the C-terminal domain of the gelatinase. ...
