PSA (prostate-specific antigen), the most useful serummarker for prostate cancer, is encoded by thehKLK3geneand is present in the serumas amixture of several molecularspecies. This work was performed to identify the hKLK3transcripts in order to determine how many proteinsresembling PSA are synthesized from thehKLK3gene andsecreted in blood. Combined Northern blotting, molecularcloninganddatabase searching showed that thehKLK3geneproduces at least 15 transcripts ranging in size from 0.7 to6.1 kb. Polysomal distribution analysis revealed that thetranscripts shorter than 3.1 kb are efficiently translated inprostate cell line....
