Humanpancreatic secretions contain twomajor trypsinogenisoforms, cationic and anionic trypsinogen, normally at aratio of 2 : 1. Pancreatitis, pancreatic cancer and chronicalcoholism lead to a characteristic reversal of the isoformratio, and anionic trypsinogen becomes the predominantzymogen secreted. To understand the biochemical conse-quences of these alterations, we recombinantly expressedand purified both human trypsinogens and documentedcharacteristics of autoactivation, autocatalytic degradationand Ca2+-dependence....
