Dehydroascorbate reductase (DHAR) reduces dehydro-ascorbate (DHA) to ascorbate with glutathione (GSH) asthe electron donor. We analyzed the reactionmechanismofspinach chloroplast DHAR, which had a much higherreaction specificity for DHA than animal enzymes, using arecombinant enzyme expressed inEscherichia coli. Kineticanalysis suggested that the reaction proceeded by abi-uni-uni-uni-ping-pong mechanism, in which binding ofDHA to the free, reduced formof the enzyme was followedbybindingofGSH....
