We purified pancreatic deoxyribonuclease I (DNase I) fromthe shark Heterodontus japonicususing three-step columnchromatography. Although its enzymatic properties resem-bled those of other vertebrateDNases I, sharkDNase I wasunique inbeingabasicprotein.Full-lengthcDNAs encodingtheDNases I of two shark species,H. japonicusandTriakisscyllia, were constructed from their total pancreatic RNAsusing RACE. Nucleotide sequence analyses revealed twostructural alterations unique to shark enzymes: substitutionof twoCys residues at positions 101 and 104 (which are wellconserved in all other vertebrate DNases I) and insertion ofan additional Thr or Asn residue into an essential Ca2+-binding site. ...
