UMP-CMP kinase catalyses an important step in thephosphorylationofUTP, CTPanddCTP. It is also involvedin the necessary phosphorylation by cellular kinases ofnucleoside analogs used in antiviral therapies. The reactivityof human UMP-CMP kinase towards natural substratesand nucleotide analogs was reexamined. The expression ofthe recombinant enzyme and conditions for stability of theenzyme were improved. Substrate inhibition was observedfor UMP and CMP at concentrations higher than 0.2 mM,but not for dCMP. The antiviral analogL-3TCMP wasfound to be an efficient substrate phosphorylated intoL-3TCDP by human UMP-CMP kinase. ...
