The ultimate step in Alzheimer’s disease Abgenerationinvolves c-secretase, which releases Abfrom its membrane-bound precursor. Asimilar presenilin-dependent proteolyticactivity is implicated in the release of theNotch intracellulardomain. We have developed a novel assay forc-secretaseactivity based on green fluorescent protein detection. Thisinvolves cotransfection of a substrate-activator based on theamyloid precursor protein or the Notch sequence and afluorescent reporter gene.
