Glycosylatedpolyenemacrolide antibiotics, as nystatins andamphotericins, are amphiphilic structures known to exertantifungal activity by disrupting the fungal cell membrane,leading to leakage of cellular materials, and cell death. Thismembrane disruption is strongly influenced by the presenceand the exact nature of the membrane sterols. The solutionstructures of five representative glycosylatedmembers, threetetraenes (pimaricin, nystatin A1and rimocidin) and twoheptaenes (candidin and vacidin A) have been calculatedusing geometric restraints derived from1H-NMR data andrandom searches of their conformational space....
