Mass spectrometric studies on the N-linked glycans ofaminopeptidase 1 from Manduca sextahave revealedunusual structures not previously observed on any insectglycoprotein. Structure elucidation of these oligosaccha-rides was carried out by high-energy collision-induceddissociation (CID) using a matrix-assisted laser desorp-tion/ionization time-of-flight/time-of-flight (MALDI-TOF/TOF) tandem mass spectrometer. These key experimentsrevealed that three out of the four N-linked glycosylationsites in this protein (Asn295, Asn623 and Asn752) areoccupied with highly fucosylated N-glycans that possessunusual difucosylated cores. ...
