SulA protein is known to be one of the physiologicalsubstrates of Lon protease, an ATP-dependent proteasefrom Escherichia coli. In this study, we investigated thecleavage speci®city of Lon protease toward SulA protein.The enzyme was shown to cleave27 peptide bonds inthe presence of ATP. Among them, six peptide bondswere cleaved preferentially in the early stage of digestion,which represented an apparently unique cleavage siteswith mainly Leu and Ser residues at the P1,andP1¢positions, respectively, and one or two 1 Gln residues inpositions P2±P5. ...
