Self-regulation of the 2-oxo acid dehydrogenase complexesduring catalysiswas studied.Radical species as sideproductsof catalysis were detected by spin trapping, lucigenin fluor-escence and ferricytochromec reduction. Studies of thecomplexes after converting the bound lipoate or FADcofactors tononfunctional derivatives indicated that radicalsare generated viaFAD. In the presence of oxygen, the 2-oxoacid, CoA-dependent production of the superoxide anionradical was detected.
