Acyl-CoA dehydrogenases and acyl-CoA oxidases are twoclosely related FAD-containing enzyme families that arepresent inmitochondriaandperoxisomes, respectively. Theycatalyze the dehydrogenation of acyl-CoA thioesters to thecorrespondingtrans-2-enoyl-CoA. This review examines thestructure of medium chain acyl-CoA dehydrogenase, asa representative of the dehydrogenase family, with respectto the catalytic mechanism and its broad chain lengthspecificity. Comparing the structures of four other acyl-CoAdehydrogenases provides further insights into the structuralbasis for the substrate specificityof eachof these enzymes. ...
