Rhodanese is a sulfurtransferase whichin vitrocatalyzes thetransfer of a sulfane sulfur fromthiosulfate to cyanide. Ionicinteractions of the prokaryotic rhodanese-like protein fromAzotobacter vinelandiiwere studied by fluorescence andNMR spectroscopy. The catalytic Cys230 residue of theenzyme was selectively labelled using [15N]Cys, and changesin1Hand15NNMRresonances onadditionof different ionswere monitored. The results clearly indicate that the sulfurtransfer is due to a specific reaction of the persulfurated Cysresidue with a sulfur acceptor such as cyanide and not to thepresence of the anions....
