Ouabain, a sodiumpump (Na+/K+-ATPase) inhibitor, hasbeen shown to act as a hormone and is possibly involved inthe pathogenesis of hypertension. The mechanismby whichouabain may act was investigated using primary cultures ofhuman umbilical artery endothelial cells (HUAECs), whichare known to express and release the vasoconstrictive hor-mone endothelin (ET-1). Fiveminutes after application, lowconcentrations of ouabain induced Ca2+oscillations andstimulated ET-1 release from endothelial cells into themedium. To investigate whether the observed effects weredue to inhibition of the sodiumpump, the effects of ouabainon the uptake of86Rb+by HUAECs were examined....
